Doc is not doctor and that was stated many times.
As for why is he working with the Maker and chosed "You" to be somewhat savior, then all we can tell is that they have similiar goal. "You" doesn't have a past, is one of the S3LFs and has only one purpose, to get more information on machine.

But why did he know that it was “YOU”... Why did Doc know this?...He never even says that he contacted the Creator directly...

@Archieart He knows "You" because it is made out S3LF that he took from the other place. The "player" (that is also the name for "you") is supposed to loop in order for nevada to not fall, which is in favor of The Maker and also gathers info about mandatus and machine, which benefits Doc.

Everything I said is in arena mode. It will require to play like 4 times through arena from fresh character with no imprints.

but why does Doc need to know information about the mandatus? Why this particular mission? Why doesn't Doc do it himself?

Don't know what you mean by particular mission, but I guess you mean arena mode. Story mode tells about fall of nevada and arena about it's backstage. If both would do same thing, then it could get messy.
Doc doesn't do it himself, because he is mastermind and those as usual don't put themselfs to danger + he can revive others, but it is not guaranted to him.
What is his target? Who knows? It wasn't stated exactly.

So. So I draw a conclusion. Doc learns about hell and other places in order to save his partners. He needs “YOU” to find out about the mandate and the Machine... well, “YOU”, in turn, does this on Doc’s orders. Now we draw conclusions; As far as I understand, all the characters, well, almost, do not know the Machine, and Doc sends “YOU” to carry out his order, as if he knows that this Nevada deity exists.
